Upon the discovery of a novel sequence, determining the next steps in its analysis is critical for understanding its biological significance. This chapter explores the importance of characterizing novel sequences, introduces key concepts and provides a comprehensive workflow for determining their biological significance. Thorough characterization of novel sequences is essential for understanding the functional, evolutionary, and structural properties of these sequences, thereby advancing our knowledge in genomics, proteomics, and molecular biology. The chapter also demonstrates real-world applications, such as the characterization of the novel coronavirus—later named SARS-CoV-2—during the 2019 pandemic. Furthermore, we highlight the key takeaways, emerging trends and future directions in the field of sequence characterization.
