  • How many DNA molecules are sequenced at the same time in one ZMW well or within one pore?

    • 1, both long read sequencing omit PCR step that limits the length of sequenced sequence therefore only one molecule is sequenced.
  • How many times one DNA molecule will be sequenced using PacBio?

    • The sequencing is circular, so the number of times DNA molecule will be sequences depends on the size of the molecule.
  • How many times one DNA molecule will be sequenced using MinION?

    • 2x, a single-stranded DNA goes through the pore while being sequenced, once reaching the herpine adaptor the other strand gets pulled into pore and gets sequenced too. These are called template and complement reads. A consensus of both is called 2D read
  • What expression subread means in PacBio terminology?

    • A full length read (polymerase read) cut into individual subreads - independent sequencing of a DNA molecule by removing adapters that made the molecule circular.
