Light-sheet switching mode

While alignment T-shirt targets allow approximate co-alignment of the light-sheets, one typically needs more precise alignment between the arms, down to 5-10 µm difference between the planes illuminated by left vs right light-sheet. To achieve this, place your test or real sample into the imaging chamber, go to 'Alignment' tab and press Run Lightsheet Switching Mode. This mode switches between L and R light-sheets dynamically while imaging image

While looking at the L\R-arm illuminated images of your sample, turn knob 3 of folding mirror M2 of the left arm and make the images between L and R as similar as possible in the FOV center. This affects (mostly) the offset of left light-sheet along Z-direction (detection optical axis, forward-backward from the user side).

If the L/R illuminated images look nearly identical at FOV center, but differ at the FOV periphery, it indicates that light sheets intersect with a non-zero angle between them. Turn left-arm linear stage 4 to change the angle of left light sheet. The linear stage affects (mostly) the angle of the light-sheet.
