Autoclave tape looks like masking tape with faint diagonal lines on it. These lines will turn black when the autoclaving process is complete, so that we know what’s sterile.
Using the autoclave machine:

Tap the screen to ensure that the machine is not in use. Place objects into autoclave-safe bins, and hold the “close door” button to shut the autoclave doors. Refer to the following sections to check which autoclave setting to use. Note that the number indicates how long the machine will run at high temperature: the actual run-time will be much longer.
We currently steal the autoclave bins from Jose Avalos’ lab.
1.1 Autoclaving glassware
Cover the tops of open containers (like beakers) with aluminum foil, and tape down with autoclave tape. For the bottles with blue lids, unscrew the blue caps slightly and secure with a piece of autoclave tape. All of the glassware that we have should be autoclaved on setting “30 gravity).
1.2 Autoclaving pipettes
Throw all the pipettes into an orange autoclaving bag, and tape it shut with autoclave tape. The pipettes that we have do not need to be dismantled in any way. Choose the “20 gravity” settings.
1.3 Autoclaving DI water
Use the 500 mL glass bottles with black tops. Fill with the millipore water only up to the 500mL line. Put the cap on loosely (don’t turn at all) and tape down the top with autoclave tape. You can do 4 bottles if you use 30-liq, or 6 bottles if you use 40-liq. Be sure to put the bottles in the autoclave box (with nothing else), and dry out the box when done if there is any standing water after the run.
