Basics
Configurations work generally by suppressing and unsupressing things. In parts you suppress different features to get different looking parts. In assemblies you can suppress parts and mates to get different arrangements of parts. You can also change dimensions and display options between configurations.
See the configurations tab of the feature manager (pane on left side of screen)
To create a new configuration simply right click in the empty space in the feature manager and click add configuration
Configurations are a great way for visualizing potential changes to a model without making a copy or permanently changing the model. See Assemblies and Changing Dimensions to see more on how to do this. The upshot is you will be able to toggle between versions quickly, have fewer (and more organized) files, and it's faster to make.
Motion
Motion can be achieved with assemblies or multibody parts using configurations. Both are better than the usual method using flexible assemblies. Flexible assemblies are terrible for performance and behave unpredictably.
Assemblies: Simply constrain the components in one position using mates in one configuration. In another configuration suppress those mates and reposition the components using different mates in another configuration.
