Initial conditions for crystallisation of CHIKV Mac were found in the BCS Screen (Molecular Dimensions). After optimisation, crystals grew reproducibly using sitting-drop vapour diffusion in MRC 3 Lens Crystallisation plates (SWISSCI) with a reservoir solution containing 0.1 M Tris, pH 7.8, 0.1 M Potassium thiocyanate, 0.1 M Sodium bromide, and 25 % (v/v) PEG Smear Broad with drops of 75 nl CHIKV Mac at 11 mg/mL, 40 nl of reservoir solution and 35 nl of CHIKV Mac crystal seed solution.
