The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a positive-sense, single-stranded RNA virus caused the devastating coronavirus disease in 2019 (COVID-19). Coronavirus (CoV) belongs to the Coronaviridae family, which comprises four genera: alpha, beta-, gamma- and delta-coronaviruses, where beta-coronaviruses can be divided into A, B, C, and D lineages [1]( J Virol, 86 (7) (2012), pp. 3995-4008). Each CoV genus shows wide host adaptability resulting in cross-species transmission [2](Bull Acad Natl Med, 205 (7) (2021), pp. 732-736). RNA-dependent RNA polymerase (RdRp) also known as nonstructural protein 12 (nsp12) is considered as one of the most potential and promising drug targets, it is essential for viral replication [3]( D.-G. Ahn, J.-K. Choi, D.R. Taylor, J.-W. Oh) and exhibits broad pan antiviral activity design [8](Front Genet, 8 (2017), p. 125).
