In 2013, Matthew Todd at the University of Sydney and GSK Tres Cantos secured funding from the GSK OpenLab Foundation for a one-year project entitled "An Open Source Hit-to-Lead Campaign in Tuberculosis Drug Discovery." This allowed for the pursuit of several promising-looking series, the first of which was Series 1, based on "Spiro" compounds that had earlier been identified by GSK as part of a phenotypic screen. Internal (non-public) research by GSK would be combined with open source research conducted before and during the period of the grant, to evaluate the potential of the series. The current status of the series is that it is parked. Despite high potency, ease of synthesis and demonstrated in vivo activity, there were toxicity issues that could not be overcome. These will be described in full in a forthcoming research publication.
