The GP49 is a variant of GP50 locomotive, equipped with a V12 version of the GP50's V16 645F3B engine. The GP49, otherwise identical to the GP50, was an improvement of the GP39-2 locomotive. Much like the GP39-2 compared to the GP40-2, the GP49 can visually be spotted on its radiator section; it has two fans instead of the standard 3 for normal V16 carbodies.

The GP49 was produced for its sole purchaser, the Alaska Railroad, in two batches: one in September 1983, and the other in May 1985, giving the unit a mere 1 & 3/4 year production life. The GP49 line only had 9 locomotives ever made, compared to 239 GP39-2s.

Two GP49s were converted over to 6 A-1-A axle locomotives for a Ferronor S.A, a Chilean railway, that purchased them from the Alaska RR. These locomotives still only retain 4 powered axles, as the one in the middle is not powered to make the locomotive's weight more widely distributed.
