The GP40 was EMD's first offering of the new turbocharged, 3000 HP 645 series V16 engine within their successful 4 axle General Purpose lineup. Like previous GP offerings, the GP40 excelled in virtually any and all uses, though most railroads preferred them as brawny-yet-affordable road freight engines, often purchasing them in far larger numbers than their SD counterpart. Between November of 1965 and December of 1971, 1,221 GP40s were produced. The GP40 replaced the prior GP35 and was replaced by the ubiquitous GP40-2 in 1972.

As with much of the 645 line, EMD offered the GP40 with a steam generator for passenger car heating, designated the GP40P. Like many passenger variants of locomotives marketed during the 1960s, though, few were built - only 13 for the CNJ. Most railroads were reluctant to invest in new passenger equipment by this time - the normal GP40 outsold the GP40P by a ratio of 600:6.
