Mouse Genotyping via Transnetyx

Overview
Mouse genotyping is handled through Transnetyx, which processes tissue samples collected on provided sample blocks. Follow this protocol to ensure accurate labeling and sample tracking.


Before You Start

  • Collect a Sample block from the Transnetyx locker (1st floor NRB, near the loading dock if not in lab).
  • Confirm you have all materials ready:
    • Metal ear tags & ear tag applicator
    • Ear hole puncher
    • Tweezers
    • Transnetyx sample block
    • Printed sample sheet
    • Disinfectant wipes or ethanol

Step 1: Set Up in Transnetyx

  1. Wean cages and update each new cage in the Transnetyx online inventory.
  2. Add weaned cages to the cart in Transnetyx.
  3. Enter the barcode from your sample block on the form.
  4. Assign animals to block positions:
    • Group littermates together.
    • Place males first (M), followed by females (F) on the same block.
  5. Print the sample sheet for lab records.

Step 2: Collect Samples

Procedure:

  1. Load a metal ear tag into the applicator.
  2. Scruff the animal with your non-dominant hand.
  3. Apply the ear tag so the number faces outward.
  4. On the opposite ear, use the punch to collect a small tissue sample.
  5. Place the sample into the corresponding well on the Transnetyx block.
  6. Record which ear tag number corresponds to each block position.
  7. Clean tools between strains to avoid contamination.

Step 3: Submit Samples

  • Deliver the completed block to the Transnetyx locker before 1:30 PM (daily pickup time).
  • Scan the green submission card (check with Mason or Gargi if unsure).
  • Place the block in the collection receptacle.
  • Restock: Grab two new sample blocks to bring back to the lab for future use.
  • In Transetyx Cart ensure all samples are correctly placed and click checkout
    • If animal is missing when going to collect samples, correct the block before checkout by omitting the sample from the plate (DO NOT leave empty wells where sample should be as this is an unnecessary cost

Tip: Keep your printed sample sheets organized by date and litter for quick reference when results are uploaded.
