DiI is used for mouse injection practice.

Recommended dilution: Prepare stock solutions of lipophilic tracers in dimethyl formamide (DMF), dimethylsulfoxide (DMSO), or ethanol at 1 to 2.5 mg/mL.

Our dilution: 25 mg/mL in 100% ethanol. They were divided into 4 tubes and marked as 10X. From one 10X 20 uL was further dissolved in 180 uL of 100% ethanol. This was marked as 1X. This 1X was sonicated to dissolve any undissolved particle of DiI. The color should be bright orange and towards transparent. All the stocks are stored at -20 freezer.

**Do not use food color for surgery practice instead of DiI as it diffuses readily on the next day and is not visible. 1:2 Evans blue might be an alternative. **
