Behavioral Recording Protocol (Bonsai)

Materials

  • Mice
  • Computer
  • Wired circle
  • 70% ethanol
  • Light
  • Camera

Protocol

  1. Open Bonsai on the computer.

  2. Ensure Bonsai loads the correct protocol for video acquisition.

    • Confirm that the Suffix setting is set to time-stamp.
  3. Prepare the video setup:

    • Clean the entire area using 70% ethanol.
    • Note: Mice can detect pheromones from the opposite gender, which may alter behavior.
  4. When saving the file, ensure the format is .avi.

    • If not saved as .avi, the video will not record.
    • Use the naming format:
      YearMonthDate–mouseName
      Example: 20250120-MouseA
  5. Start recording and adjust the camera focus on the bottom view.

    • The side camera is already auto focused.
    • Focus controls are located on the right-hand side.
  6. Pause the video and delete the test recording.

    • The next video should now be properly focused.
    • Prepare a Post-it note with the mouse name and date.
  7. Show the post-it note to the camera for labeling, then place the mouse into the chamber.

    • Record for 30 minutes. This timer is started once the door to the suite is closed.
    • Close the top portion of the chamber.
  8. After 30 minutes, remove the mouse from the chamber and stop the recording.

    • Ensure the video files are saved to the correct behavioral computer directory.
