Materials:
- Plates.
- pH strip.

| Yeast extract (Ref. 212750, BD) | 5 g |
| Peptone (Ref. 211684, BD) | 10 g |
| Dibasic potassium phosphate, K2HPO4 (Ref. P8281 Sigma-Aldrich) | 1 g |
| Glucose (Ref. G8270, Sigma-Aldrich) | 10 g |
| Agar (Ref. 214010, BD) | 20 g |
| Distilled Water | 1 L |
Method:
- Prepare the YPPD medium (table 1).
The pH should be 7,5-8. Verify with a pH strip.
- Autoclave for 15 min at 121ºC.
- Wait till the temperature is low enough to pour the medium into the plates.
Under the hood. Plates work better if they are poured thin 15-20 mL/ 10 cm Ø plates.
- Let them cool till solidification.
Can be left overnight.
- Invert the plates and dry for 10 minutes in the heater.
- Put them all inside the plastic wrapper.
- Store at 4ºC.
