Materials:

  • Ward’s cereal grass powder (cat. no. 944 V 8602, Ward’s Natural Science).
  • Distilled water.
  • Sterile deionised water.
  • Paper filters (Whatman #1).
  • Filters 0,22 µm.

Method:

  1. Prepare the Cereal Grass medium:
  • Add 2.5 mg/ mL Ward’s cereal grass powder to distilled water.

  • Boil for 15 minutes.

  • Allow to cool and filter through paper filter, followed by 0,22 µm membrane filter.

Perform under sterile conditions to facilitate downstream DNA and RNA isolation protocols.
Should produce clear straw-coloured liquid.

  1. Add equal portions of sterile deionised water and Cerela Grass medium.

Dear Inaki, you can use SP Media to cultivate them but I found filtered and autoclaved seawater to be better. Just shake the culture gently, then poor about 2-5 mL to the new flask (under the sterile bench). I add one autoclaved grain of wheat or quinoa as carbon source for the bacteria and store them in darkness at 8°C. I did place the recipe on the choanowiki page under protocols...
