Materials:
- 950 mL autoclaved & membrane filtered (pore size 0.2 µm) natural seawater.
Natural seawater can be replaced with high quality synthetic seawater made with Milli-Q water.
- 50 mL autoclaved distilled water.
- Organic enrichment (proteose peptone/yeast extract mixture) for Seawater medium.

Proteose peptone | 0,4 g |
Yeast extract | 0,08 g |
Distilled water | 100 mL |
Method:
- Add the natural seawater (950 mL) and the distilled water (50mL) together when they are cool.
- Prepare the yeast extract mixture (table).
- Dispense the organic aliquots into 2 mL Bijou bottles, autoclave and store them in a cool place.
For standard growth medium add 40 µL of peptone/yeast extract to 10 mL of seawater.
