Marine Broth 2216

Materials:

  • 37,4 g of Marine Broth powder (Ref. Marine Broth 2216, BD-Difco).
  • 1L of distilled water.
  • Filters 0,22 µm.

Method:

  1. Suspend 37,4 g of Marine Broth powder in 1 L of distilled water.
  2. Heat with frequent agitation and boil for 1 minute.

This step is to dissolve the Marine Broth powder, so it's not a problem if it doesn't boil for 1 min.

  1. Autoclave for 15 minutes at 121ºC.
  2. Allow to cool and filter through 0,22µm membrane filter.
  3. Store at 4ºC.

Marine Agar 01-291

Materials:

  • 55,1 g of Marine Agar powder (Ref. 01-291, Scharlau).
  • 1L of distilled water.
  • Plates.

Method:

  1. Suspend 55,1 g of Marine Agar powder in 1 L of distilled water.
  2. Bring it to boil.
  3. Autoclave for 15 minutes at 121ºC.
  4. Wait till the temperature is low enough to pour the medium into the plates.

Under the hood.

  1. Let them cool till solidification.

Can be left to overnight.

  1. Invert the plates and dry for 10 minutes in the heater.
  2. Put them all inside a plastic wrapper and store at 4ºC.

Culture list:

  • Abeoforma whisleri: 17ºC/Dark.
  • Corallochytrium limacisporum: 23ºC.
  • Creolimax fragantissima: 12ºC.
  • Pigoraptor chileana: 17ºC.
  • Pirum gemmata: 17ºC/Dark.
  • Sphaeroforma arctica: 12ºC
