Marine Broth 2216
Materials:
- 37,4 g of Marine Broth powder (Ref. Marine Broth 2216, BD-Difco).
- 1L of distilled water.
- Filters 0,22 µm.
Method:
- Suspend 37,4 g of Marine Broth powder in 1 L of distilled water.
- Heat with frequent agitation and boil for 1 minute.
This step is to dissolve the Marine Broth powder, so it's not a problem if it doesn't boil for 1 min.
- Autoclave for 15 minutes at 121ºC.
- Allow to cool and filter through 0,22µm membrane filter.
- Store at 4ºC.
Marine Agar 01-291
Materials:
- 55,1 g of Marine Agar powder (Ref. 01-291, Scharlau).
- 1L of distilled water.
- Plates.
Method:
- Suspend 55,1 g of Marine Agar powder in 1 L of distilled water.
- Bring it to boil.
- Autoclave for 15 minutes at 121ºC.
- Wait till the temperature is low enough to pour the medium into the plates.
Under the hood.
- Let them cool till solidification.
Can be left to overnight.
- Invert the plates and dry for 10 minutes in the heater.
- Put them all inside a plastic wrapper and store at 4ºC.
Culture list:
- Abeoforma whisleri: 17ºC/Dark.
- Corallochytrium limacisporum: 23ºC.
- Creolimax fragantissima: 12ºC.
- Pigoraptor chileana: 17ºC.
- Pirum gemmata: 17ºC/Dark.
- Sphaeroforma arctica: 12ºC
