Materials:

  • 2,5 g of Cerophyll (Ward's Natural Science Establishment, Inc., P.O. Box 92912, Rochester, NY 14692).
  • 1,1 L of Distilled water.
  • 0,5 g of Na2HPO4.
  • Filter paper (Whatman #1).
  • Agar for Klebsiella pneumoniae:
Agar for Klebsiella pneumoniae Amounts
Agar 20 g
Yeast extract 4 g
Glucose 0,16 g
Distilled water 800 mL
  • Equivalent to cerophyll: Grass Media Culture (Dairy Goat Nutrition P.O. Box 22363, Kansas City, MO 64113).

  • Similar to cerophyll: Cereal Leaf Product no. C-7141 (Sigma Chemical Company, P.O. Box 14508, St. Louis, MO 63178).

Method:

  1. Prepare solution 1: Rye grass Cerophyll.
  • Add 2,5 g of cerophyll to 1L of distilled water.
  • Boil for 5 minutes.
  • Add 100 mL distilled water to compensate for evaporation.
  • Filter through filter paper (Whatman #1).
  • Add 0,5 g Na2HPO4.
  • Adjust the pH to ~ 7,0.
  • Autoclave for 15 minutes at 121ºC.
  1. Prepare solution 2: Agar for Klebsiella pneumoniae (table).
  • Dispense in 5 mL amounts.
  • Autoclave for 25 minutes at 121ºC.
  • Slant.
  1. Bacterium, grown on solution 2, is added to solution 1 and incubated at 30ºC for 24 hours prior to inoculation with Paramecium.
