1. Adding a new Animation

Done through the menu item 'Edit > Add New Animation to Sprite Sheet', pressing 'Alt + A', or by pressing the 'Add' button under the Animations section of the side bar.

2. Adding a new Sprite

After clicking on the animation you wish to ass the sprite to in the Animations List click 'Edit > Add New Sprite to Animation', pressing 'Alt + S', or clicking the 'Add' button under the Sprites section of the side bar.

3. Saving your Sprite Sheet

Ensuring that your Sprite Sheet has at least one Animation in it and at least one Animation has a Sprite in it you can click 'File > Save', or pressing 'Ctrl + S'. You will then be prompted to select a location and file name for your exported sheet. All files will be created in the location you select. A FileName.XML file will be created along with a FileName_##.PNG file for each animation in your sprite sheet, where FileName is the file name you give it and ## being the number of that animation in your Sprite Sheet.

4. Opening an Existing Sprite Sheet

So long as the .XML and all it's accompanying .PNG files are kept together you can open the saved Sprite Sheet without having to pass around the image files used to build the Sprite Sheet. All necessary image data will be pulled from the .PNG files themselves. This is done by clicking 'File > Open', or pressing 'Ctrl + O' and selecting the .XML file to open.
